Ryan, There shouldn't be a problem. The only reason you would experience a complication is if there is activity on your network that would utilize one of the roles, like making a change to the Schema, or a change to an account that would need to replicate to an NT4 BDC. If you are going to reboot after hours, and if you can keep developers, admins, etc, from modifying AD, you'll be just fine. Then you can split out the roles later.
