I would go into the Network Connections and select the network adapter on the server. On the Microsoft File and Print item, select properties. And make sure the settings are optimizes for file and print sharing. Next you could pull up perfmon and see what the network usage is for the box, and number of SMB sessions that are on the box. Also you could be seeing MBLAST related issues. NIC settings on the server could be a problem, if the settings are set to auto negotiate, and the switch port is either not set or set differently. Finally Disk subsystems are very important when you have more than one person connecting to the drive. IDE is optimized for 1 or 2 concurrent users.
