Yes, it will undo the proxy settings if you un-check the policy in IE
Maintenance. The only exception to this may be if you're using
preference mode IE Maintenance policies, which are only supposed to run
once, and thus probably wouldn't be smart enough to undo themselves.

I want to test our proxy server. I can enable the proxy settings for IE
in a GPO by checking 'enable proxy settings'. 
My question is if I need to remove the proxy settings, will unchecking
the 'enable proxy setting' disable the proxy settings in IE?
