If I were doing this, I would not just uncheck the quota options, I would
actually check the "use mailstore" option, or whatever it is called. It's the
topmost checkbox. It is better for you to set your limits at the Store level.
 
In any case, here's what you asked for:

Move all the 600 users to an OU (in our script, the OU is called myOU, change
that to match the name of the OU where you have the users)
Change 'LDAP://myDC.MyDomain.com/OU=myOU,DC=myDomain,DC=com' to reflect your
setup.
Then change the other "LDAP://myDC.MyDomain.com"; to also reflect the name of
one of your GCs
 
Then save the file as a .vbs file and double-click on it.

Yes, the storage limit are not through store level policy. They have been set
as manual.
I am very novice as for as scripting is concern.If you do not mind could you
please write the script for uncheck Prohibit send at and prohibit at box for
me with steps how I would run it.
Can I run this script on a OU because I am planning to move these users to a
new OU.

        I have 1000 users in my AD in a single OU. We have set the mailbox
