Canyflyer, you would want to render a project- or part of the project in form of the 
workarea as you only then see the real effects.This means that with some tricky 
effects and transitions you can see the effect without rendering it but not with the 
same quality as you would when rendered. Also sometimes when you have too many effects 
on your clip your computer might give up and you have to render. I personally always 
use breaks to render my timeline.
So in short rendering gives you the full quality view of all your effects.

I have AP v6.5 and I was told {which turned out to be right} that all you have to do 
is go to project settings>key frame and rendering> and check the real time video box 
and then your able to preview {by pressing enter} the timeline with all the effect. 
Now why would anyone want to render their project if they didn't have to?

that depends on the version of Premiere you are on. In Pro (not sure about 6 and 6.5) 
you can preview without rendering. It should work automatically but you can check in 
projectsettings under video. You can also render a certain part by adjusting the work 
area on the timeline and then go into render> workarea.
