Actually, it is not the scratch disk locations to check for “conformed” files, it is the Media Cache. You can find this under Edit > Preferences > Media. Note there are two entries, one for “files” and the other “Database”. Both are necessary. Just by way of explanation, it is my understanding that these files are shared between Premiere Pro, After Effects, Media encoder, Audition, Encore, or whatever so that as you move from application to application, conforming does not have to be redone within each of them. This was done to save time. AFAIK the setup is the same between CS3 and later versions, so I can’t suggest a reason you have problems with CS5 only. But perhaps one way to troubleshoot is to begin by cleaning the cache (there’s a button to do that but you can also simply use Windows Explorer to simply delete the files). It is safe to do this but recognize older projects will of necessity be re-conformed. Next, create a test project and import one file, wait for conformance to complete, then examine the file Media Cache files’ name and date/time stamps. Close your project, reopen and confirm the files are persistent across reloads. On a following day, verify the existence of the same files in the cache - prior to starting Premiere Pro, and if they are missing then that would explain the problem and you’d look to find out why. Otherwise, reopen the project and confirm it doesn’t re-conform. If it re-conforms even though the same cache files are present, then I don’t know what to do next.
