If this is truly just one file, I think your only choices will be a 300+ GB disk pool on the TSM server or LTO-2 tape drive. I'm using LTO drives (LTO-1) and get a rock-solid 18 MB/sec (63 GB/hr). For just a single file, we're talking about more than 4 hours for the backup.
I don't know of anything that will run faster than LTO-2 that would have the capacity you need, except a big disk array. What is the nightly backup window for this file, and what type of system does it sit on? Would client-side compression be viable?
