Which type of installation you have done for Exchange 2010 ?
Are you in DAG mode or no ?
If you are in DAG mode your install is not properly configured
Verify too you opt files.
I'm configuring a LAN-Free Solution with a DAG actually.
This is my configuration for information :
1 node for DAG (EX2K10PROD_DAG)
6 node for Exchange (DSSxxxxxxxx_EXC for each node of DAG/DAC)
All Exchange's node was granted to save for DAG's node

Backups worked yesterday and today are failing with a very cryptic message:

ACN5350E An unknown Exchange API error has occurred.
Nothing is in the Event log, TDPEXC.LOG about what the api error is. Doing a
QUERY EXCHANGE gives a little more information, but still nothing in any
event log to indicate what exactly the problem is
