I'd like to help you figure this out. Below is me "thinking out loud".... If you re-deploy (or update the web.config or something), then the application cache is flushed along with the with the AjaxPro handlers. And when you run an AjaxPro request, the page lifecycle is not executed. I wonder if it has something to do with the AjaxSettingsSectionHandler.Create() not being fired and not in cache.
