Left hand threaded nuts are normally marked.  The left handed lug nuts
have a mark in the middle of each point of the nut.  The Spica nut may be
marked the same way, assuming it is left hand thread.
    You can also examine the exposed threads.  Left handed threads should be
obvious. However, the threads on the Spica pump are way down in the engine
compartment.

I just want to make sure before I start wrenching on my Spica pump pulley
the direction of the threads for the main nut holding the toothed pulley on.

Some time ago I was removing the pulley on the smog pump (also run in the
same direction as the Spica pump) and found out after a lot of wrenching that
the nut was reverse threaded. So I'd like to ask before I try the Spica
pulley.
