Assuming your amavisd-new user is 'vscan', try this first: What does
su vscan -c 'pyzor ping'
say?

Then run:
su vscan -c 'pyzor discover'

and try
su vscan -c 'pyzor ping'
again.

If your amavis user does not have shell access, then you can directly
edit the 'servers' file that pyzor uses. This file should be located in the
.pyzor directory of the amavis user's home directory, somewhere similar
to /var/amavis/.pyzor and at this time it should contain:
66.250.40.33:24441

Like Mark says, if Pyzor can't find its server, or it does not get
reply packets back, it may produce these symptoms. If you can't get a good
pyzor ping, and you can't determine the reason why, then you would
have to disable pyzor.

You probably know this stuff, but just in case.
