If you are expecting the user to be able to access the desktop application when they are on 3G or otherwise not on the same WiFi network, port forwarding is required > The person with the application running on their desktop > may not know how to set up port forwarding in their router. Their router may not even support port forwarding. If you are limiting your interest to when the user is on the same WiFi network as the desktop application, port forwarding is not needed, but somehow the user will need to know the IP address of the desktop and be able to provide it to the phone.
