No, you should just get an error message. But if that worries you you
could copy your AUTOEXEC.BAT and CONFIG.SYS to a boot floppy, and
edit out the lines concerned in the floppy copy. Then boot up from the
floppy disk.

But you *will* have to reset your CMOS setup. In fact on bootup your
computer's self-test should show an error and swing you into the setup
screen. Then you set the cylinders etc for the second hard drive
according to what is on the printed label on the new hard disk and save
the new setup.
