Go the the folder where you installed email engine and look at the emaildaemon.properties file. There should be a parameter in it which tells how often to recahce the config changes. You need to wait till that time or just restart email service. If windows the default install directory is: C:\Program Files\BMC Software\ARSystem\AREmail Are you using servergroup by chance?
