Epub is generated by passing the docbook generated by the docbook backend through the xslt stylesheets, it does not use the HTML5 backend. There are experimental xslt stylesheets for epub3 that includes video and audio IIUC so if you would need to set your stylesheet options for xsltproc to reference those.
Also the docbook backend does not support video, so you would have to use passthrough or make your own customisation for it to generate the docbook videoobject entity.
