all you need is installing an LDAP server on windows and sync
it with the hMailServer accounts, at that point you'll be able to use
LDAP from ASSP to validate addresses, otherwise you may just
setup ASSP to use VRFY/EXPN so avoiding the need for the flat
file and at the same time having the addresses always "in sync"
to do so, you'll have to enable the HELP/VRFY/EXPN commands
on hMailServer and then configure your local domains list to look
this way

domain1.tld=>serverIP:port
domain2.tld=>serverIP:port
domain3.tld=>serverIP:port
domain4.tld=>serverIP:port

where "domainX.tld" is the domain name, serverIP:port are the IP
and the port on which the server used to VRFY addresses for that
domain is listening
