Definitely.

It works like this: -

1       Some one sends me a request for information
2       I spend time and effort composing a reply, which I send to the same
email address that requested the information
3       I get a challenge, requesting a response
4       That's rude. I ignore it, and never speak to that person again.

As one of the links quoted says, NDRs of this form are themselves bulk
unsolicited mail = spam!

If you use Challenge-Response as an "anti-spam" measure, since the vast
majority of spam or virus originated mail is from forged addresses, you may
well find yourself listed as a spammer. Stay away from the entire concept,
would be my advice.
