We've seen something similar, and it was an IMail thing.

Check in your SMTP service configuration - I'll bet you've got a check
in "Auto Deny Possible Hack Attempts" which sounds like a good idea
unless all your smtp traffic is coming from the same box. Any possible
hack attempts are always identified as coming from ASSP, and then the
IMail server stops accepting any connections from ASSP.

I've unchecked that box, made sure the ASSP IP address is removed from
the deny list under the "Control Access (SMTP)" area. Since we funnel
all SMTP traffic through ASSP, I'm considering changing the rule in that
area from "allow all except" to "deny all except". Right now, that's
basically the way it is anyway because of firewall rules, but...
