It would be exten => 101,1,Dial(SIP/[EMAIL PROTECTED])

If Asterisk does not find a [host.domain.com] entry in sip.conf it will dial by hostname/IP address. Only settings in sip.conf [general] section will be used. People do this with Asterisk all the time. You are not trying to do anything special.
