I just set this same configuration up yesterday: 2T100P cards in 1
asterisk box, and yes, it does work..
/etc/zaptel.conf:
span=1,1,0,esf,b8zs
span=2,1,0,esf,b8zs
bchan=1-23,25-47
dchan=24,48
