Honestly python can do what you asked. Although, do not ever forget that it is on you to code your game/app, not on the language. Most of the famous languages are capable of handling audio games well enough, as audio games have much less calculations and computing compared to video games. That is my scripting language of choice.
The rest of the post is about C#.
I know that BPCSharedComponent written by munawar the developer of TDV is publically available and supports key/sound handling. If you, however, don't feel like using that library, you can always use directX features and wrap them to your liking. Yes. Things are more complicated in C# compared to python, but again, C# is used in Unity, up to that extent that all Unity scripts are in C#. And the game you mentioned is also coded in C#. So it is your choice. It's just about work. You can work harder and longer and get enough knowledge of C# and get cool things out of it, or choose python and use third party libraries like pygame and code your game based on them. Syntax also matters. If you ask me, check both languages and see which syntax sounds better to you.
