One thing you might consider is an ABX or ABXY blind test.  It works
like this - suppose you think you can hear a difference between A and
B.  To test that, your friend flips a coin to choose if X=A or X=B. 
Then she or he plays A and B and X when you request, and you have to
decide if A is X or B is X.  ABXY is just the same, but where you
decide if A is X and B is Y or if A is Y and B is X (slightly easier
for the listener; slightly harder for the friend to keep straight!). 
If you can get it correctly say, 9/10 times (each trial beginning with
a fresh coin flip), that's good evidence you can in fact tell the
difference.  I would recommend closing your eyes or turning your back
so you don't see your friend, to avoid any visual cues, or even better
set it up so that she or he doesn't know which SB is which. 

By the way, the media player foobar has a nice plugin which will set up
ABXY for you automatically given, say, two audio files encoded with two
different lossy formats.

Rememeber - randomly choosing will be correct 50% of the time, so you
really have to do at least 10 tests, and get essentially all of them
right, to be confident you can hear a difference.
