Follow-up Comment #1:
This is documented: if your filename (in particular on
Windows operating systems) contains a colon, the
full form of the -U option is mandatory, i.e. all four
subcomponents (<memory area>:<operation>:<filename>:<format>)
must be present.
This is the only way to disambiguate between the colon that
is part of the filename, and the colons that divide the
various subcomponents of the -U option.
The shortened form(s) of the -U option only work with
filenames that don't contain colons.
(Besides, %temp% must be resolved by the shell/command
processor calling AVRDUDE.)
