See page 110, Using Markers, in the BBEdit User Manual. In a nutshell, you can select a range of text (or just use the current cursor position) and choose the Set Marker command from Mark popup menu in the Navigation Bar. Alternatively, you can Control-click on the selected text and choose Set Marker from the contextual menu. To assign a keyboard shortcut to this command (or any other menu command), go to Preferences -> Menus. The Set Marker command can be found under the Navigation Bar -> Markers collection.
