First, try moving the app to your Applications folder. Then, when you run, does the Quicksilver icon appear in the dock? Click that to 'activate it'. The normal hotkey is ⌃-space, if that's what you use for Spotlight then you can change the Quicksilver hotkey in the Prefs:
