No reboot necessary. Clear it with
/etc/rc.d/init.d/iptables clear
then run again
/etc/rc.d/init.d/iptables start
I have also use manually "iptables -F" (-F = --flush) and all have
cleared.

-F, --flush [chain]
Flush the selected chain (all the chains in the table if none
is given). This is equivalent to deleting all the rules one by
one.
