One of the best ways to block a site from being accessed is to put an entry 
in the HOSTS file.  The default HOSTS file in Windows usually only has one 
entry, which is:
127.0.0.1   localhost

This is a required entry and must not be removed.  For each site that you 
would want to block, you would add entry such as:
127.0.0.1  www.anysite.com
127.0.0.1  anysite.com

All of these entries would be inserted below the default entry listed above.

I have been using this HOSTS file for several years now and I wouldn't surf 
without it.  They update the file on a regular basis, but I only download 
the latest version about every six months.
Additionally, I place other entries in the HOSTS file, to block my daughters 
computer from visiting sites that we don't want her using.
The MVP HOSTS file combined with the Bluecoat K9 Web Protection 
http://www1.k9webprotection.com is an excellent way to control which web 
sites can be accessed from a given computer.
