I don't think Speakup supports usb synths.
But I don't own a hardware synth my self so I'm not sure.
I know Speakup supports serial port synths though and I think it's possible to get a usb synth to work by using a usb to serial converter.
But don't quote me on that!
I use software speech with Speakup when I use Linux.

I doubt Orca (the GNOME screen reader) supports usb synths either.
In fact I don't use Orca at all.

But a good place to get answers is the Speakup mailing list.

I can dig it up if you are interested, but a google search for "Orca screen reader" should help as well :)
