The folks who sell means to "game" the SAT tests probably have memorization of the words most likely to be on the test as part of their plan. So, in that sense, memorizing a small subset of the words in the English language could help. But, there is no real way to sit down and memorize 25,000 words to go from a 25k vocabulary to a 50k vocabulary. Indeed, if one looks at the people with the largest vocabularies, one finds that they are extensive readers of writers that use a large vocabulary. That's the natural way to learn the meaning of words. That's why a survey of reading material gives a good measure of one's vocabulary.
