Yes, you will at least need to logout and login after the installation.
Rebooting shouldn't be necessary, though.

You are probably the first one to try LilyPond on Vista, so I'm afraid
you cannot expect too much help here. However, the first of your
error printouts is printed by LilyPond itself and the last ones, which
come from the embedded Scheme interpreter, are probably consequences
of the first one. What you can do to assist the debugging even further is
to run
lilypond.exe --verbose
and send the corresponding output to the mailing list.
