In answers to your questions:

Yes, use a framework. You will all in all be better off for it.
Cake is probably the best balance of ease of use and power.
Prototype is supported by cake, so if you are just starting with ajax,
that's the route of least resistance.
For job interviews PHP is probably enough. You can usually learn the
framework on the job if they use one. But, if you say you also know a
framework, it can only help.

If you take the time to learn aframework, you will almost certainly
become a better developer.

Start with the cake manual, play with the tutorials, read the lists,
put in the hours. You will be happy you did.
