Well, there is a preference setting that will automatically launch the file after the file transfer is completed. Navigation -> Downloads -> Automatically open downloaded files
This option is turned off by default.
I know, that's why I want to have a faster way to toogle ist. I usually leave it off, but when I'm listing to amazon realaudio-sample autostart would be great (or even better would be inline playing support which I wasn't able to configure yet :-( ).

Not sure what is meant here. Open and Remove are both in the toolbar and in the contextual menu when clicking on a completed dl file.
Thats why I put the " here, one button which opens the file AND removes the entry would be useful. Usually you only need to open a downloaded file once, so there's no use to leave it in the download-manager.
