Dear Shivesh,

Is your solvent content or  Mathew's coefficients are reasonable with your current  model?

Did you check the packing?  Is there any void among the molecules?

R-factor and R-free is seems to be reasonable with your resolution and also you mentioned that  you have not seen any density for missing part.  If you have missing domain in your model and you have not  accounted that portion in the structure factor calculation that might be reflected  in your R-factor and R-free.

So most likely that ordered domain might be crystallized and flexible domain might be cleaved off during crystallization,  even though you used the full length protein.

Mass spectra on your crystal might give you more information.

Dear all,
At present we have 2.4A resolution.the protein is supposed to have two domains separated by a 8-residues linker region with 3 glycines which we have checked with DisEMBL.So,one domain is can be flexible.We have done the mass spectroscopy of the protein solution itself and we are trying to check with the crystals also.There is no tag in the protein.We kept two acetate molecules and 13 water molecules and it has two bound calciums.
