I received several replies to my original question, then a little hiatus, then I made the Summary. Following the summary, some more contributions to my original question came. So, I would like to add to the summary: - Do not care about tag names, common sense is to care not to have a long extenstion. Consider carboxypeptidases to remove C-terminal tags, eventual heterogeneity might not hurt. - Consider BioRad new kit for producing tag-less very pure protein. Great with proteins of compromised solubility or low expression. - Tags effects highly dependent on the protein, etc. Again, might or might not help crystallization. Two experiences with c-Myc tags , no crystals with or without, other approach needed (the cost of the resin to be considered).
