Hi All, I was wondering if anyone had troubles with PES (Polyethersulfone) ultrafiltration membrane filters (Vivaspin, Amicon,...) using 5 mM DTT containing buffer. It seems whether or not it is possible for DTT to react with sulfone groups of the membrane and in this way to "crosslink" my cystein rich protein (I can't recover after ultrafiltration neither in the retentate vial nor in the filtrate vial). I was wondering if using regenerated cellulose ultrafiltration membrane (now relatively difficult to find in labs) might be a solution to overcome this problem.
