Hello All, I have purified recombinant Aspergillus CYP51 from E.coli. But it seems that there is protein there but it is white and not red. No hemeprotein is present as seen in spectral studies or by eye. The spectral studies were done ~ 1mg/ml concentration. I did include 5-aminolevulinic acid during induction and the purified protein shows the presence of 6X-His tag by in-vision His tag gel stain (Invitrogen).
Any thoughts as to why heme was not incorporated? Is it possible to reconstitute the protein-Heme complex? I appreciate your help in advance. Thanks Hari
