I have been trying to answer the following questions:
1) In the context of optical imaging, how is a protein crystallography 
reconstruction validated? How meaningful is this perspective of protein 
crystallography (optical imaging)?
2) In transition to single particles (single unit cells), what additional 
factors should be considered?
I am not a chemist or a crystallographer, and the question for me is the 
applicability/accuracy of the scattering model(s). I will be grateful if you 
could comment on my below-mentioned answers to these questions. For a quick 
check, I would suggest "Abstract + Introduction + Tables 1 & 2 + Conclusions".
