Job description Dr. Julien's laboratory at The Hospital for Sick Children Research Institute in Toronto, Canada is looking for a Research Fellow to join our Structural Biology team. The successful candidate will participate in the characterization of B cell glycoprotein receptors by using a combination of biochemical, biophysical, immunological and structural techniques. We are looking for a driven and enthusiastic scientist with the passion to apply his/her knowledge and creativity towards the structure/function study of biologically-important targets.
Roles & Responsibilities * Design and optimize constructs for expression in mammalian cells; * Biophysically characterize purified glycoproteins for their Tm, Tagg and polydispersity with the objective to improve crystallizability; * Perform ITC and BLI binding assays between glycoproteins and purified antibodies to improve binding thermodynamics and kinetics; * Determine X-ray crystal structures of glycoproteins and glycoprotein-antibody complexes; * Use Electron Microscopy as a target screening tool and to determine the molecular structures of complexes of interest. Desired Skills and Experience * Structural Biologist with a PhD in Biochemistry, Immunology, Molecular Biology, or related field; * Strong knowledge and practical experience with the structural biology of therapeutic proteins, especially glycoproteins and antibodies; * Experience in molecular biology, mammalian protein expression, purification and biophysical characterization (Octet, Optim, ITC, SEC-MALS) is a plus; * Demonstrated ability to participate in collaborative scientific projects * Organization skills, efficiency and meticulous record keeping About Us Dr. Julien's laboratory is interested in the structure/function study of the B cell receptor and critical co-receptors on the B cell surface. A better understanding of the structure of these B cell surface molecules, as well as those that propagate intracellular signals will allow careful modulation of their function and will be instrumental for vaccine adjuvant research looking to elicit protective antibodies against variable pathogens. It will also provide unique sites to specifically target deregulated B cells in childhood acute lymphoblastic leukemia (B-ALL), as well as in autoimmune diseases such as Type-1 diabetes, rheumatoid arthritis (RA) and systemic lupus erythematosus (SLE).
