Hi All, I am seeking to purify a protein of interest that has an approximate size of 160 kDa. The construct includes a variety of purification tags, including an HRV 3C site for proteolytic cleavage, a GGS linker that provides flexibility, a Twin-Strep tag for affinity purification, a short flexible linker for improved folding, and a 3×FLAG tag, also included for ease of detection and purification. Given this complex structure, I am seeking the most suitable resin to use for purifying the protein from HEK293 cells.
In my research, I came across Strep-Tactin® XT Affinity Resin, which seems promising. However, I would appreciate any insights on whether this is the ideal choice or if other options might offer better results for purifying my protein construct. Additionally, if any individuals have experience utilizing a specific resin from a particular company, I would appreciate your insights.
