It's certainly possible, and depending on your server configuration you may
not have to change anything at all. However, I would recommend that you
actually secure access to the directory instead (or in addition). This is
typically a tiny bit harder, but is a much better solution.

You can do this by using your web server's functionality. You can create a
separate virtual server that isn't publicly accessible, you can use web
server authentication to check access to the directory, you can use SSL to
prevent passwords from being gathered from network traffic analysis.
