Hello John, I have done a lot of websites that utilize IIS Integrated Authentication. The largest one as of yesterday was standing at 2.98 Terabytes.
You will need to have a couple of ports open on your firewall that will allow your webserver access to the AD servers. You will need TCP port 135, and TCP/UDP port 389 for LDAP. Here is a link for the TCP Port numbers used by Microsoft. http://support.microsoft.com/default.aspx?scid=kb;en-us;179442 Here is a good link to aid in trouble-shooting. http://support.microsoft.com/default.aspx?scid=kb;en-us;816103 Hope this points you in the right direction.
