Why are you going to use MS SMTP again?  We send emails to large amounts of
users as well, and have had no problems since we switched over to iMS lite
(free) with a dedicated mail server.  We are using CF 6.1 standard and have
not seen noticeable delays.  

If you need better performance, you can upgrade to one of their higher end
versions (they go by # of threads, the free one is 5 threads, and I believe
the pay versions start at 25 threads and go up from there).  

iMS can also send out email by itself.  We are using it with a dedicated
mail server box because we already had one and we wish to use that box for
what it's meant for instead of causing more resource drains on the CF
server, but it would probably work well without a dedicated server as well.

There is nothing specific about MS SMTP, the optimizations 
Macromedia recommends for optimizing CF email performance apply 
equally to MS SMTP. (Except for the number of threads.) Check the 
Macromedia site for the whitepaper.
