There are a couple of possibilities, and it (somewhat) depends upon
what version of Windows you're running.

If you replace a single-core CPU with a dual-core CPU, Windows XP and
Win2K3S should detect the change and change the system type from
uniprocessor to multiprocessor automatically, and load the appropriate
driver.

For Windows 2000, go into the Hardware Device Manager, find the
"uniprocessor PC" entry, right click, select "update driver", click on
"display list of known devices" and find the multiprocessor PC driver.
 You'll have to reboot.

For the 2nd option, I would recommend Ghosting your system beforehand,
just in case.

Knowing a bit more about your OS and hardware would be helpful.  You
