Just the opposite.
Thanks to your suggestion, we solved the issue.
I was going crazy in IIS, trying to find what configuration was off.

But after your suggestion, I just checked the actual directory permissions, and 
somehow the Read permission had been set to "Deny" for the entire wwwroot.

You deserve a bottle of champagne, for your assistance.
