We faced the same problem a month or two back.

CF is coping nicely with 300MB files. No tweaks of the JVM. Can't say for
sure but I don't remember what we set the large upload values in CF Admin
to, but it's been coping nicely.

One thing I would recommend, is to make it easy to swap the upload pages,
both the form and the action pages, with something else should things not
turn out too well.

We have a Flex file uploader to make things nicer for the user. I'd
certainly recommend giving them some sort of feedback as the upload
progresses.
