Not directly within the <VirtualHost>.  If you want to do that you'll
have to put it within a <Directory> directive:

<VirtualHost *:80>
    ServerName  dev.local
    DocumentRoot "C:/dev/sites/default"

    <Directory "C:/dev/sites/default/playground/404">
        RewriteEngine on
        RewriteCond %{REQUEST_FILENAME} !-f
        RewriteRule ^(.*)?$ 404.cfm?page=%{REQUEST_URI} [QSA,L] 
    </Directory>
</VirtualHost>
