What have you defined in clamd.conf's LocalSocket, TCPSocket, TCPAddr?

You don't have to post the values, just make sure that there is something
reasonable set for those options.

The message you got means that freshclam could not make a connection on the TCP
socket, it could be that clamd is not running or if it is running it is not
listening to that port or type of socket.
