I don't know of a strict definition, but larger companies (Fortune 1000, say) tend to have stricter standards on the applications they run in terms of reliability and support, as well as features, configurability and scalability. An application, say a CMS, suitable for a 10-person office will probably not handle the needs of a 30,000-person company. For example, look at Interwoven TeamSite <http://interwoven.com/products/content_management/index.html>: definitely an "enterprise" app, fully stocked with all applicable bells and whistles, which means installation, configuration and customization is definitely non-trivial. Which means in practice that you'll have to pay their professional services people until you develop in-house expertise.
