I'm not a OSGEO4W user, but I've no objections to modifying shapely/geos.py so that it can find the right GEOS DLL in that case. Patches gratefully accepted ;)
BTW, 1.2.1 win32 installers are uploaded to PyPI and http://gispython.org/dist.
