The daisy chained drives are 80GB, 160gb and 500GB containing 56GB,
140GB and 121 GB of data respectively. Is that OK?
What's the 500GB limit related to ... a proportion of the size of the
1TB backup drive?
Does the Time machine 1 TB backup drive have to be empty or can it have
some data on it also? (I assume any data on it could not be part of
the Time Machine backup....)

I want to install a 1 TB SATA II drive internally in a Leopard G5 on the
2nd onboard SATA controller and use it as the target drive for Time
Machine to back up the main drive and 3 HFS+ daisy chained external
firewire drives.
Is that possible and easily so?
Should work with ease. This assumes the 4 drives being backed up are
small (no more than 500GB total).
